Our study concerns the reliability of techniques for obtaining accurate and quantitative full-length 16S rRNA amplicon sequencing data on bacterial community structure with MinION (Oxford Nanopore Technologies). We conducted 1) Evaluate the accuracy of taxonomic assignment by filtering based on sequence length. 2) Compare five PCR conditions using three independent mock microbial community standard DNAs. 3) Establish appropriate, standardized, optimal PCR conditions among the trials investigated in 2). We sequenced two mock communities and six environmental samples with Illumina Miseq for comparison.
