Probing the effects of oxidative stress on the membrane binding of alpha-synulcein

Neutron reflectometry will be used to characterise the cellular membrane binding interaction of alpha-synuclein under conditions mimicking oxidative stress. Alpha-synuclein is the main component of Lewy bodies the characteristic marker of Parkinson's disease. Aggregation of alpha-synuclein is thought to lead to the formation of Lewy bodies. Oxidative stress has been linked to neurodegenerative diseases including Parkinson's disease and therefore the membrane binding interaction of alpha-synuclein under oxidative stress is of interest to investigate whether this causes aggregation of the protein and changes to the membrane binding interaction. Tethered lipid bilayer membranes will be used to investigate alpha-synulcein membrane binding after oxidation with Fe(II) and ascorbate and with a synthetic "oxidised" phospholipid incorporated.
