Effect of Phosphatidylserine lipids on Tissue Factor structure and function in lipid membranes

Tissue Factor (TF) is the membrane protein responsible for the initiation of the blood coagulation process [1]. In case of a vessel injury, the TF is exposed to the blood stream and initiates a series of cascade reactions the first of which being the formation of a protein complex with the coagulation factors FVIIa (TF/FVIIa) and FXa (TF/FVIIa/FXa). No experimental data has so far been published on the structure of TF or TF/FVIIa/FXa in lipid membranes. Recently we developed a new protocol for fast and robust preparation of lipid bilayers with membrane proteins, which we propose here to use for investigating TF. The proposed experiment will provide the needed data to finalise a manuscript already in preparation. A PhD student is involved in this project; this additional experiment will provide fundamental experimental data for his thesis (thesis submission deadline in September).
