Understanding the regulatory roles of small RNAs (sRNAs) in Mycobacterium marinum is crucial for elucidating its pathogenesis. Here, we present transcriptome profiles of M. marinum strains with deletions and completions of sRNA B11. Through RNA sequencing analysis, we identified significant alterations in gene expression patterns between the B11-deleted and completed strains. Overall design: Wild-type (WT + pCtr), B11-deleted (?B11 + pCtr), and B11-deletion complemented with over-expressed B11 (?B11 + pPb11-B11) strains of Mycobacterium marinum were cultivated in 7H9-OADC medium until reaching the early exponential phase (OD600 ~ 1.0). At this stage, B11 expression in WT strains stabilizes. Bacterial cultures (4 mL) were mixed with 0.2 volumes of stop solution (95% ethanol and 5% phenol) and immediately frozen in liquid nitrogen. Total RNA was isolated using the Trizol method.
