RNA-Seq analysis for overexpression of SdeA

To analyze the function of sdeA in A. fumigatus, we overexpressed the sdeA gene in the A. fumigatus wild-type strain A1160 using the gpdA (A. nidulans glyceraldehyde-3-phosphate dehydrogenase) promoter. To explore the mechanisms by which a high-level expression of SdeA mediates fungal growth, a transcriptome sequencing (RNA-seq)-based approach was performed. Two groups of samples were collected. In group 1, conidia from the OEsdeA-5 mutant were cultured on MM for 16 h before RNA extraction. In group 2, conidia of wild type that were cultured on MM for 16 h served as the control.
