Transcriptional responses of V. parahaemolyticus to Zn limitation.

RNA sequencing analysis of the transcriptome of V. parahaemolyticus RIMD2210633 strain grown under Zn-deficient (LB pretreated with 35 µM TPEN) and Zn-replete conditions (LB pretreated with 35 µM TPEN along with 500 µM Zn). Overall design: The RIMD2210633 strain was grown in LB broth to the early-exponential phase (OD600 of ~0.7). Subsequently, two aliquots (1 mL) of the culture were removed and bacterial cells were collected by centrifugation at 6 000 rpm for 5 min. The two bacterial pellets were resuspended with 1 mL LB pretreated with eiher 35 µM TPEN alone or 35 µM TPEN along with 500 µM Zn. After incubation for 15 min at 37°C, total RNA was isolated and subjected to RNA sequencing analysis. Biological triplicate samples obtained from three independent experiments were used.
